SpringWorks Therapeutics, Inc. is a biopharmaceutical company dedicated to identifying, developing, and commercializing innovative therapies for patient populations grappling with rare diseases and various forms of cancer, often those with significant unmet medical needs. The company's most advanced experimental treatment is nirogacestat, an orally administered small molecule that functions as a gamma secretase inhibitor. This drug is currently progressing through Phase III clinical trials for the management of desmoid tumors. Another key asset in their pipeline is mirdametinib, an oral small molecule designed to inhibit MEK. It is being evaluated in Phase 2b clinical studies for neurofibromatosis type 1-associated plexiform neurofibromas (NF1-PN), and concurrently in Phase 1/2a trials for the same condition. Furthermore, mirdametinib is part of a combination regimen with lifirafenib, currently in Phase 1b/2 clinical assessment for patients with advanced or treatment-resistant solid tumors. The company is also exploring mirdametinib both as a standalone agent and in various combination strategies for solid tumors. Additionally, SpringWorks is advancing BGB-3245, an oral, selective small molecule inhibitor targeting both monomeric and dimeric forms of activating BRAF mutations, which is in Phase I development. SpringWorks has forged strategic alliances, including collaborations with BeiGene, Ltd., GlaxoSmithKline LLC, and Allogene, focusing on combination therapeutic approaches involving nirogacestat and mirdametinib. They have also secured license agreements, notably with Pfizer Inc. for nirogacestat and mirdametinib. Additional licensing deals include an arrangement with Katholieke Universiteit Leuven and the Flanders Institute for Biotechnology for a collection of novel small molecule inhibitors of the TEA Domain, and a separate agreement with Dana-Farber Cancer Institute for a portfolio of novel small molecule inhibitors of Epidermal Growth Factor Receptor. Moreover, the company maintains clinical collaboration agreements with several entities, such as Janssen Biotech, Inc., Precision BioSciences, Inc., Seagen Inc., and AbbVie, Inc. Founded in 2017, SpringWorks Therapeutics, Inc. maintains its corporate headquarters in Stamford, Connecticut.
